My garden is covered in ground elder which I have been trying to tackle by digging over, removing as much as possible and spraying with weedkiller for over a year now. I want to lay a new lawn. Is there a membrane I can use under the new turf that will prevent the ground elder coming up through it?

I suspect that isn't going to work - your turf would sit like a carpet on top of the membrane, wihout a firm grip, and in dry weather would not be able to draw moisture from below the membrane, so most likely would dry out and die. I am only theorising mind, as I have never heard of the idea before.
I would not have dug AND sprayed, as digging chops teh roots up into tiny bits and each bit becomes an independent weed. I would just spot spray, and spot spray again 10 days later, and keep on until no more ground elder appears, and then prepare the ground for turfing.
my allotment was covered with the stuff two years ago and I used round up ultra 3000 on it and after repeat applications it is now clear but the cost was two bottles at £40.00 a bottle,mind my allotment measures 100 feet long and 30 feet wide.
I only used two bottles because the first application i used a strong measure and this just burned the leaves so no liquid was able to soak down into the roots.
if you are going to use round up only mix what it says on the bottle.
IT WILL CLEAR / KILL GROUND ELDER.
I agree digging will only make it worse, I had this problem in a garden last year & had to resort to spraying ( which is always a last resort) only a tiny bit has come back this year which I will spot spray.
I also wouldn't lay a lawn on membrane, I'd wait for ground elder to die first,
